Ounce you have deburred the flash hole, and uniformed the primer pocket on new brass.
This step doesn't have to be done again correct?

Correctamundo. You only need (read: should) clean the primer pocket after firing with your uniformer. You'll probably remove a very small amount of brass, but not to worry, as it won't affect anything in regard to weakening the case.
